What is 5-Amino-1MQ?

5-Amino-1MQ (5-amino-1-methylquinolinium) is a small organic molecule, not a peptide. It is a quaternary quinolinium cation with a molecular weight of approximately 159 Da, studied as a selective inhibitor of the enzyme nicotinamide N-methyltransferase (NNMT).

Research background

The compound was developed at the University of Texas Medical Branch as part of screening quinolinium analogues as NNMT inhibitors. Results in cellular and mouse metabolic models were published in 2018.

Mechanism and scientific context

NNMT transfers a methyl group from S-adenosylmethionine to nicotinamide, producing 1-methylnicotinamide. It thereby links vitamin B3 metabolism (and, indirectly, NAD+) to cellular methylation metabolism. NNMT inhibition is studied as a way of influencing nicotinamide availability and cellular energetics in adipocytes.
