What is kisspeptin?

Kisspeptins are a family of peptides produced by cleavage of the protein encoded by the KISS1 gene. The main forms are kisspeptin-54 (also known as metastin), kisspeptin-14, -13 and -10. All share the same C-terminal decapeptide, which is sufficient for receptor activation.

Research background

The KISS1 gene was discovered in 1996 in Hershey, Pennsylvania, as a suppressor of melanoma metastasis, which gave it its name. Only in 2003 did research into the genetics of puberty show that mutations in its receptor disrupt the reproductive axis, moving kisspeptin to the centre of reproductive neuroendocrinology.

Mechanism and scientific context

Kisspeptin activates the KISS1R receptor (formerly GPR54), a G-protein-coupled receptor expressed on hypothalamic GnRH neurons. Activation stimulates GnRH release and, consequently, LH and FSH release from the pituitary. Kisspeptin is therefore considered a key upstream control point of the reproductive axis.
