What is PE-22-28?

PE-22-28 is a short synthetic seven-amino-acid peptide (GVSWGLR) derived from the larger peptide spadin. Spadin is a fragment of the propeptide of sortilin, a receptor involved in intracellular protein trafficking.

Research background

Spadin was described around 2010 by Jean Mazella's research group in France as a natural inhibitor of the TREK-1 potassium channel. PE-22-28 was later developed as a shortened analogue intended to retain activity with greater stability.

Mechanism and scientific context

TREK-1 is a potassium channel of the K2P family that regulates neuronal excitability. In animal models its inhibition is associated with changes in behaviour and hippocampal neurogenesis. The body of work is small and mostly comes from the same research group.
