What is semaglutide?
Semaglutide is a 31-amino-acid analogue of the human hormone GLP-1 with a molecular weight of approximately 4.1 kDa. Compared with native GLP-1(7-37) it contains three key modifications: aminoisobutyric acid at position 8, arginine at position 34, and a C18 fatty diacid chain attached to lysine via a linker.
Research background
Semaglutide was developed by Novo Nordisk as a successor to earlier GLP-1 analogues. Regulated pharmaceutical preparations containing semaglutide are approved medicines with their own documentation, which does not apply to research materials.
Mechanism and scientific context
Native GLP-1 is degraded by the enzyme DPP-4 within minutes. Aminoisobutyric acid at position 8 protects the molecule from this enzyme, and the fatty chain enables albumin binding. Together these modifications extend the half-life from a few minutes to about one week — a textbook example of rational peptide design. Semaglutide is a selective GLP-1 receptor agonist.
