What is bremelanotide?
Bremelanotide is a synthetic cyclic heptapeptide with the sequence Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-OH and a molecular weight of approximately 1025 Da. It is structurally almost identical to Melanotan 2, differing only by a free carboxyl group at the C-terminus instead of an amide.
Research background
Bremelanotide emerged from the line of melanocortin analogue research begun at the University of Arizona and was later developed by Palatin Technologies. A regulated pharmaceutical preparation is approved for a narrowly defined indication and has its own documentation.
Mechanism and scientific context
Bremelanotide is a melanocortin receptor agonist with prominent activity at MC4R, a receptor expressed in the central nervous system. The small structural difference from Melanotan 2 is enough for the two molecules' profiles to be studied separately, making them an interesting pair for structure–selectivity research.
