What is SS-31?

SS-31 (elamipretide) is a synthetic tetrapeptide with the sequence D-Arg-Dmt-Lys-Phe-NH2, where Dmt denotes 2′,6′-dimethyltyrosine. The molecular weight is approximately 640 Da. Its alternating arrangement of aromatic and positively charged amino acids allows it to cross cell membranes.

Research background

The SS designation comes from Hazel Szeto and Peter Schiller, who developed this family of peptides. The molecule later entered clinical development as elamipretide for rare mitochondrial diseases.

Mechanism and scientific context

SS-31 accumulates selectively at the inner mitochondrial membrane, where it binds cardiolipin, a phospholipid essential for organising respiratory chain complexes. In experimental models it is associated with stabilisation of cristae structure, efficiency of oxidative phosphorylation and reduced generation of reactive oxygen species.
