What is tirzepatide?
Tirzepatide is a synthetic 39-amino-acid peptide whose sequence is based on the hormone GIP, with an attached C20 fatty diacid chain. The molecular weight is approximately 4.8 kDa. Albumin binding extends the molecule's half-life.
Research background
Tirzepatide was developed by Eli Lilly as the first dual GIP and GLP-1 receptor agonist. Regulated pharmaceutical preparations containing tirzepatide are approved medicines with their own documentation, which does not apply to research materials.
Mechanism and scientific context
Tirzepatide activates the GIP and GLP-1 receptors and has been described in pharmacological studies as an “imbalanced” agonist with more pronounced activity at the GIP receptor. At the GLP-1 receptor it shows a bias towards cAMP signalling over β-arrestin recruitment, a subject of active receptor pharmacology research.
